Quantification of intraoperative tumor-cell spillage of neuroblastoma and rhabdomyosarkoma by FACS and RT-PCR

In pediatric patients with advanced solid tumors the minimal residual disease i.e. the persistence of vital tumor cells in the body after medical and surgical therapy is of utmost importance for the prognosis. It is conceivable that the inevitable manipulations of the tumor during the resection by the surgeon results in a tumor-cell spillage in the adjacent tissue and blood and lymphatic vessels. While this is an accepted fact for tumors of the adult population, there is no single study investigating this issue for tumors in childhood. In this clinical study we look for disseminated tumor cells in tissue fluid and peripheral blood before, during and after surgery for advanced neuroblastoma stage IV and mestastasizing alveolar rhabdomyosarcoma. Highly sensitive analytical methods like FACS and RT-PCR are applied which are provided by a cooperation with the Pediatric Clinic ( Dr. Lode). The proof of tumor cell spillage during the operation would be of great significance for surgical technique and perioperative medical treatment.
